Resistance of carbapenemase-producing Klebsiella pneumoniae isolated from patients with orthopedic infection

Objective. To evaluate the resistance rate and production of carbapenemases in Klebsiella pneumoniae with phenotypic resistance to carbapenems isolated from patients with orthopedic infection. Materials and Methods. The materials for the study were tissue samples, aspirates and removed orthopedic devices of patients with orthopedic infection at the Vreden Russian Research Institute of Traumatology and Orthopedics, between 2017 and 2019. K. pneumoniae strains were identified in Microlatest by iEMS ReaderMF. These strains were tested for susceptibility to 15 antimicrobial agents by disk diffusion methods, as described by the EUCAST.

The rates of mutations associated with macrolide resistance in Mycoplasma genitalium among patients with non-gonococcal sexually transmitted infections in Smolensk and Tula

Objective. In this study, we examined the prevalence of macrolide-resistant M. genitalium in two Russian cities, Smolensk and Tula, between 2013 and 2017. Materials and Methods. DNA’s samples were isolated from urethral and cervicovaginal swabs using primary screening and tested for macrolide resistance-associated mutations by real-time PCR. This technology makes it possible to identify any nucleotide substitutions in the 23S rRNA M. genitalium gene at positions 2058, 2059, 2611 M.

Detection of acquired carbapenemase genes among Acinetobacter baumannii isolated from blood culture in patients with hematological malignancies

Objective. To evaluate the prevalence of acquired carbapenemase genes among A. baumannii isolates collected from blood culture in patients with hematological malignancies. Materials and Methods. This prospective multicenter study included A. baumannii isolated from blood culture in hematological patients in 7 Russian hospitals (2003–2015). Minimum inhibitory concentrations (MICs) of meropenem and imipenem were determined by broth microdilution method (CLSI, 2017). All A. baumannii isolates were tested by multiplex real-time PCR for presence of acquired carbapenemase genes (blaOXA-23-like, blaOXA-24⁄40-like, blaOXA-58-like, blaNDM-like, blaVIM-like and blaIMP-like).

Susceptibility testing of Candida glabrata clinical strains to echinocandins using SensititreTM YeastOneTM system

Objective. To determine susceptibility of C. glabrata isolates to anidulafungin, caspofungin and micafungin using the SensititreTM YeastOneTM system. Materials and Methods. C. glabrata isolates were taken prospectively from clinical specimens or from strains collections in the participating sites. Susceptibility determination was performed using SensititreTM YeastOneTM (YO10 panel) according to the manufacturer’s guidance, and results were interpreted with M27-A3 CLSI guidelines. Susceptibility of C. glabrata to fluconazole was also determined in order to assess possible correlations of echinocandins and fluconazole minimal inhibitory concentrations (MICs) in resistant strains.
