Local experience of real-time PCR implementation into microbiological monitoring programs

Objective. To study level and structure of microbial colonization in cancer patients before admission to a surgery department using molecular methods. Materials and Methods. The presence of DNA of methicillin-resistant Staphylococcus aureus, methicillinresistant coagulase-negative staphylococci, Candida albicans/C. glabrata/C. krusei, Acinetobacter baumannii in the clinical specimens and the presence of antimicrobial resistance genes in cultures were detected by multiplex real-time PCR. A total of 741 clinical samples and 313 cultures were studied.

Clinical and epidemiological features of infections caused by HHV-6A and HHV-6B

Objective. To determine the detection rates of HHV-6A and HHV-6B compared to other herpes viruses in children of different age groups. Materials and Methods. A total of 128 patients with symptoms of respiratory viral infections (RVI) aged from 1 to 16 years and 101 otherwise healthy children. A comprehensive examination of the patients, including ELISA, indirect immunofluorescence assay, rapid culture method, PCR, and determination of nucleotide sequences of HHV-6 was performed.

To evaluate epidemiological efficacy of the 10-valent pneumococcal conjugated vaccine (PCV10) as assessed by the number of episodes of respiratory tract and ENT diseases and by the number of antibiotic courses prescribed for those diseases in children under 5 years of age in Barnaul. Materials and Methods. At the end of 2016, a total of 312 children under 60 months of age were enrolled into a retrospective cohort epidemiological study and divided into 2 cohorts (156 children each).

Etiology of community-acquired pneumonia in the military personnel

Objective. To assess the etiology of community-acquired pneumonia (CAP) in the army recruits using a polymerase chain reaction (PCR) assay. Materials and Methods. Sputum, blood, bronchoalveolar lavage samples and oropharyngeal swabs collected from 255 hospitalized army recruits with radiographically confirmed diagnosis of CAP were tested by PCR assay. The comparator group included 270 otherwise healthy recruits. Detection of Streptococcus pneumoniae, Haemophilus influenzaе, Mycoplasma pneumoniae, Chlamydophila pneumoniae, Chlamydophila psittaci, Legionella pneumophila, adenoviruses, Herpes simplex virus I/II, cytomegalovirus was performed by PCR using commercial AmpliSens kits and GenPak DNA PCR test.

Objective. To assess the possibility of standard disk application as an antibiotic source for determination of the minimal inhibitory concentrations (MICs) by serial microdilution method. Materials and Methods. A simple method of working solutions preparation using standard paper discs as an antibiotics source available in most microbiological laboratories is proposed.
